Pharmacokinetics of Hydroxysafflor Yellow A in Rats

A HPLC method for the determination of hydroxysafflor yellow A in rats was established. The DiamonsilTMC18 column was used with mobile phase of methanol-acetonitrile-0.45% H3PO4 solution (32.5∶2.5∶65), at detectionwavelength of 402nm. The calibration curve was linear in concentration range of 0.1～200mg/ml. The detection limitwas 10ng/ml. The average recoveries of high, medium and low concentration were 90.8%, 88.2% and 82.2%, respectively.The RSDs of intra- and inter-day were less than 7%. The pharmacokinetics parameters for doses of 2, 6, 18mg/kg wereAUC0→4h 5.34, 13.50, 35.25mg·h·ml-1, V(c) 0.1199, 0.1290, 0.1242 L/kg, Cl(s) 0.3743, 0.4445, 0.5107 ml/min,respectively.
