Identificazione di specie in prodotti lattiero-caseari con tecnica PCR-RFLP

In this work the AA. report the set up of a PCR-RFLP method to obtain a clear and reliable identification of the four principal species that interest the dairy industry (Bos, Ovis, Capra, Bubalus) in preparations which have been heat-treated, matured or processed with other technological procedures. This method is based on the comparative analysis of 63 sequences of the cytb gene belonging to all the four species taken into account. This analysis allowed to identify two regions whose sequences were used to design two universal primers which were subsequently utilized in the PCR amplification of a part of cytb gene from pure or mixed dairy products; the amplified sequences contain sites recognized by specific restriction enzymes that can generate species-specific restriction profiles.
