Promotion of Neuronal Guidance Growth by Aminated Graphene Oxide Via Netrin-1 / Deleted in Colorectal Cancer Signaling

2020
Promotion of neuronal outgrowth and synapse formation is a key step for nerve regeneration and depends strongly on the injured local expression of axonal guidance cue Netrin-1. Aminated graphene oxide (NH2-GO) displays electrical properties that make it appealing for neuronal regenerative medicine, yet its interaction with primary cultured cortical and spinal cord neurons has been scarcely investigated. Here, we report a molecular mechanism of NH2-GO on axonal guidance growth in central nervous system. NH2-GO concentrations (2μg/mL, 4μg/mL) were selected considering that cell viability increased significantly by CCK-8 assay. Notably, NH2-GO exhibits increased reactivity and can react with many other compounds, thereby improving the dispersibility of graphitization amination and strengthening the effect of the composite material. More interestingly, the use of NH2-GO (2μg/mL, 4μg/mL) up-regulation of Netrin-1 and its receptor DCC (Deleted in Colorectal Cancer) in cortical and spinal cord neurons by immunof...
