Effect of extract from prunella vulgaris L on Jurkat cell apoptosis

Aim:To investigate the apoptosis-inducing activity and the related mechanism of the extract from prunella vulgaris L on human T-lymphoma leukemia Jurkat cell line.Methods:The inhibition ratio of extract from prunella vulgaris L on Jurkat cells was tested by MTT assay.Apoptosis of Jurkat cells was observed by agarose electrophoresis.The expressions of apoptosis protein Bcl-2 and Bax were detected by immunocytochemistry.Results:The extract from prunella vulgaris L could remarkably inhibit the proliferation of Jurkat cells,and the IC50 was(53.59±3.10) mg/L.The agarose electrophoresis showed typical DNA ladder;and the immunocytochemistry showed that the expression of Bcl-2 protein decreased((12.31±1.21)% vs(40.76±1.64)%) and the expression of Bax protein increased((18.14±0.39)% vs(4.77±0.16)%).Conclusion:The extract from prunella vulgaris L can inhibit proliferation and induce apoptosis of Jurkat cells,which probably realized by down-adjusting the expression of Bcl-2 and up-adjusting the expresion of Bax.
