Construction of wide-type p53 gene vaccine

Objective:To construct a wide type p53 gene vaccine which has the therapeutic activity.Methods:We get the intact wide type p53 gene from human peripheral lymphocyte total mRNA using RT PCR,then cloned it to TA vector and link to pCMV after gene amplification.pCMV p53 expression vectors were transfected in DU145 using lipofectamine and the protein was detected by immunohistochemistry.Results:p53 gene can express stability in DU145.Conclusion:We constructed a p53 gene vaccine what can express stability in Du145 cell line.
