The invention provides a quadrupole guide rod radio frequency power supply circuit of a mass spectrometer. A CPLD signal source circuit outputs a frequency and amplitude adjustable high-frequency square wave signal; then, a drive circuit carries out amplification on the high-frequency square wave signal to generate a drive signal; and a radio frequency transformer circuit generates a sine wave radio frequency signal according to the drive signal and a feedback signal output by a feedback circuit and outputs the signal to a guide rod, wherein the feedback signal is a signal for carrying out retroegulation on the sine wave radio frequency signal when the sine wave radio frequency signal is beyond a preset range, that is, the quadrupole guide rod radio frequency power supply circuit of the mass spectrometer can set frequency and amplitude of the high-frequency square wave signals through software, and adjust the sine wave radio frequency signal through the feedback signal, so that frequency and voltage of the sine wave radio frequency signal are controlled to be suitable for ions in different quality ranges.
