The biological characteristics of stem cells transplanted into glioma models

2009
Objective After embryo neural stem cells (NSCs) and bone marrow stromal cells (BMSCs) were cultured and amplified in vitro, and transplanted into glioma from rats,to observe the chan-ges of biological characteristics of NSCs and BMSCs and neurospengioma. Methods NSCs and BMSCs were isolated from the hippocampus of the rat embryos (fetal age of 15 days) and cultured in vitro.Estab-lish glioma models and in the 3w and 4w selected 5 rats randomly to enforce MRI detection.Draw the ma-terials after being fixed at the different time, HE staining and BrdU immunohistochemistry.Observe the Cell livability and the impact on glioma. Results NSCs and BMSCs that transplanted into glioma models were able to survive. The average survival time of the BMSCs group, NSCs transplantation group and the control group models respectively is 4.03 weeks, 4.28 weeks, 3.88 weeks; the average diameter of the glioma when growed three weeks and four weeks respectively is (6.4 mm/7.6 mm,5.4 mm/6.0 mm,7.2 mm/8.0 mm).BrdU staining showed that two types of cells are directed targeting to the glioma ceils.Conclu-sion NSCs can inhibit the growth of glioma cells, extending the survival time of rats.BMSCs have no sig-nificant inhibition on the growth of giioma cells. The two types of cells are directed targeting to the glioma cells, and they can be used as biological carrier transfering target genes to treat glioma.
