Analysis of microsatellite information in EST resource of leafhopper.

【Objective】EST sequence information for leafhopper was analyzed to provide references for setting up molecular marker system, constructing genetic mapping and identifying associated injuring genes. 【Method】Based on the four types of leafhopper subspecies EST sequences downloaded from NCBI data library, the SSR motifs and corresponding SSR primers were found through the bioinformatics method. 【Result】A total of 11044 EST sequences with the total length of 5866 kb was obtained. According to design parameters, 1946 EST sequences accounting for 17.6% of the total EST se-quences were screened, which contained 2230 SSR motifs. Their corresponding SSR primers (average distance of 2.63 kb)were also obtained. In addition, trinucleotide was the major type of motifs in 2-5 nucleotide repeats, which accounted for 77.3%. Particularly, AAT motif was the most abundant among trinucleotide repeats and accounted for 40.7%. After that, AG motif was the largest among dinucleotide repeats and accounted for 39.1%. 【Conclusion】A total of 11044 leafhopper EST se-quences which contained a large number of SSR motifs was analyzed. As a result, the trinucleotide motif type was the largest one and 2230 pairs of SSR primer were obtained.
