Carcinogenesis – the novel hypothesis and new targets for anti-cancer treatment

Presented article suggests the novel hypothesis of carcinogenesis, where the key moment for all types of carcinogenesis (biological, physical, chemical) has been discussed. The thorough theoretical analysis of the mechanisms of malignant transformation of cells after influence of carcinogens has been presented. Hypothesis highlights importance of mitochondrial electron transport chain alterations, or disorders in major energetic apparatus of the cell, cytochrome structure disorders, increase in mitochondrial pH, disorders of enzyme functioning, in particular pyruvate dehydrogenase leading to anaerobic glycolysis, homocysteine accumulation and its further penetration in cell nucleus, and DNA mutations in the mechanism of cell malignant transformation. According to the suggested hypothesis the new targets for cancer therapy has been emphasized.
