Effect of the Carbohydrate Source on Improvement of Adventitious Regeneration in Plum (Prunus Domestica L.)

ABSTRACTThe influence of the carbohydrate source on the regeneration of different type of explants as leaves and stem segments from in vitro plants, cotyledons and hypocotils isolated from immature and mature stored seeds of the plum cultivars Stanley and Kyustendilska sinya was studied. Regeneration media composed of MS salts and vitamins with addition of 7.5 μM TDZ and 2.5 μM IBA were enriched with sorbitol or sucrose alone, or in combination. As optimal carbohydrate source for all type of explants was defined the combination of 101.5 mM sorbitol and 29.2 mM sucrose promoting the highest morphogenic reply.
