Effects of proteasome inhibitor on apoptosis of hepatocellular cancer cell line BEL7402

Objective: To explore the role of proteasome inhibitor MG-132 on apoptosis of human hepatocellular cancer cell line BEL7402.Method: MTT assay was used to calculate the cell inhibition rate,AnnexinⅤ/PI staining was used to determine the apoptosis rate,electron microscope was used to observe the morphologic change,agarose gel electrophoresis was carried out to detect the DNA fragmentation,and colorimetric assay was used to measure the activity of caspase 3.Result: MG-132 inhibited BEL-7402 cell growth significantly,which acted further with the raised concentration and prolonged the treatment of MG-132.Typic ultrastructure of apoptosis cells was found by electron microscope after the cells were treated with MG-132.Agarose electrophoresis revealed marked ladder and activated caspase 3 in the treatment group.Conclusion: MG-132 inhibits the proliferation of BEL-7402 cell and promotes its apoptosis.
