Studies on the Genetic Stability of Regenerated Plants from Siraitia grosvenorii Shoot-tips after Cryopreservation

[Objective] The research aimed to examine the genetic stability of regenerated plants from Siraitia grosvenorii shoot-tips after cryopreservation by vitrification method.[Method] Plants before cryopreservation and those regenerated from Siraitia grosvenorii shoot-tips after cryopreservation by vitrification were examined by peroxidase and esterase isoenzyme analysis.The number of chromosomes was counted and the molecule markers of random amplified polymorphic DNA(RAPD) was made.[Result] Before and after cryopreservation,there were 9 bands on POD isoenzyme patterns and there were 8 bands on EST isoenzyme patterns,with the same mobility.There were no variations before and after cryopreservation at peroxidase and esterase isozyme patterns,the number of chromosomes and RAPD patterns.[Conclusion] Cryopreservation of S.grosvenorii germplasm resources by vitrification was safe and stable.
