Su principal aportacion se considera que fue la tecnica denominada de fundoplicatura para el tratamiento de la hernia hiatal, aunque realizo otras que no han tenido tanta relevancia cientifica. Tambien ha sido recordado como el cirujano que trato quirurgicamente a Albert Einstein de su aneurisma de aorta abdominal, mediante una tecnica que no fue efectiva de recubrimiento con pelicula de celofan. Rudolf Nissen inicio su formacion y trabajo en Alemania vinculandose como ayudante al prestigioso cirujano Sauerbruch, para posteriormente trasladarse a Turquia potenciando el desarrollo de la cirugia de este pais, emigrar mas tarde a Estados Unidos trabajando en inicialmente en Boston y mas tarde en New York y retornar a Europa exactamente a Basilea para culminar su carrera profesional.
