The expression of clusterin in canine renal tubular epithelial cells treated with different concentrations of calcium oxalate

Objective
To investigate the relationship between clusterin expression and development of renal calculi.
Methods
Detect the expression level of clusterin in canine renal tubular epithelial cells (MDCK) and supernatant fluid after treated with different concentrations of calcium oxalate by Western blotting method and enzyme linked immunosorbent assay (ELISA) method.
Results
Clusterin expressed in both control cells and cells treated with calcium oxalate solution, but obviously stronger in treated cells. The clusterin concentration in supernatant fluid of cells treated with different concentration (300, 500, 900 μg/ml) was (241.33±59.28), (247.67±22.48), (270.67±43.50) pg/ml, respectively. Which was significant higher than that in control, (78.67±39.80) pg/ml (P 0.05).
Conclusion
Clusterin expression increased in renal tubular epithelial cells treated with calcium oxalate, which indicate it may be involved in the formation of kidney stones.
