Biological property of a rope-shaped scaffold braided by silk-poly (L-lactic acid)-polyglycolic acid fiber

Objective To evaluate mechanical property and biological property of a rope-shaped scaffold braided by silk-poly (L-lactic acid) (PLA)-polyglycolic acid (PGA) fiber,and investigate the feasibility of fabricating tissue-engineered ligament by using such scaffold.Methods Firstly,a hybrid rope-shaped scaffold containing monofilament 7200 was prepared by twisting a mixture of silkworm silk fibroin,PLA,and PGA fiber filament with a mass ratio of 1∶1∶1.The biomechanical property of the abovementioned silk-PLA-PGA scaffold was measured by using the electric universal testing machine.Secondly,the biological property of the silk-PLA-PGA braided scaffold materials was assessed according to the relevant experiments,including cvtotoxicitv test,acute systematic toxicity test,intracutaneous irritation test,pyrogen test,and muscle implant test.The experimental results were analvzed and assessed by criterion.Results The appearance of the scaffold braided by silk-PLA-PGA was a strong obdurability with a diamter of 2.7 mm.The scaffold had excellent mechanical property,good histocompatibility,no cytotoxicity,no acute systematic toxicity,no skin stimulation,and no pyrogen reaction.Conclusion The scaffold has good biomechanical property and biocompatibility.
