Method for constructing long fragment sequencing library

Provided in the present invention is a method for constructing a long fragment sequencing library. The method comprises the following steps: 1) preparing a 5184-well plate containing a single-stranded DNA; 2) placing the single-stranded DNA in equal amounts into each well on the 5184-well plate; 3) placing an entire genome amplification reaction system into each well of the 5184-well plate processed in step 2) and reacting the same; 4) placing a fragmented reaction solution into each well of the 5184-well plate processed by the step 3) and reacting the same; 5) adding different tag sequences to nucleic acid molecules in each well of the 5184-well plate processed in step 4) to obtain the sequencing library.
