Change of Hydrolase Activities in Transgenic Anti-sense Thioredoxin s Gene Wheat in the Process of After-ripening

In order to exploring a effective way for improving wheat seed storage,we detected the seed respiration speed and the activities of thioredoxin h,α-amylase,β-amylase,debranching enzyme and protease in transgenic line and non-transgenic cultivar 'Shengkang No.1' by enzyme activity assay during seed after-ripening stage.The results indicated that the activities of thioredoxin h,α-amylase,β-amylase,debranching enzyme and protease were significantly lower and the seed respiration velocities decreased distinctly in transgenic line.During after-ripening 0~30 d,the activities of thioredoxin h,α-amylase,β-amylase,debranching enzyme,protease and seed respiration velocities were 16.07%,31.64%,6.44%,21.63%,27.67% and 52.90% lower than that of non-transgenic seeds,respectively.The above result demonstrated that foreign trxs gene interfered the expression of the endogenous gene,decreased Trxh activity,seed respiration velocities and hydrolyses activities and retarded converting velocities of reserve materials.Therefore,the seed storability was improved.
