RP-HPLC determination for the content and related substances of triamcinolone acetonide acetate and chloramphenicol solution.

To establish an RP-HPLC method for the determination of the content and related substances of triamcinolone acetonide and chloramphenicol solution. Dionex C18 (250 mm × 4.6 mm, 5 m) column was adopted, the mobile phase consisted of 0.1 mol L–1 - heptanesulfonic acid sodium salt (including 0.1 mol L–1 1-heptanesulfonic acid sodium salt, 0.5 mol L–1 potassium dihydrogen and 0.5% triethylamine, adjust pH = 2.5 with phosphoric acid)(A) - methanol (B) with gradient elution (0 min(66% A) → 20 min (66% A) → 30 min (35% A) → 49 min (35% A) → 50 min (66% A) → 60 min (66% A)) at the flow rate of 1.0 mL min–1; the detection wavelength was 277 nm during 0-20 min and 230 nm during 20-60 min. The linear response ranges of chloramphenicol, triamcinolone acetonide acetate and naphazoline hydrochloride were 30.375-3.000 mg mL–1 (r = 0.9990), 0.0127-0.1016 mg mL–1 (r = 0.9989) and 0.0262-0.2096 mg mL–1 (r=0.9992). The result of determination is accurate and specific. This method is suitable for determination of the content and related substances of Triamcinolone Acetonide Acetate and Chloramphenicol solution.
