Effects of the Gekko Swinhonis anti-neoplasm active component on the proliferation, migration and apoptosis of HepG2 cells

2012
Aim To investigate the effects of the Gekko Swinhonis anti-neoplasm active component(GSAAC) on the proliferation, migration and apoptosis of HepG2 cells. Methods HepG2 cells were treated with GSAAC.Cell proliferation and migration were determined by MTT and Transwell assay after treatment. The expression of PCNA was detected by immunohistochemistry.Hochest33342 fluorescence staining and TUNEL staining were used to observe the morphological changes of HepG2 cells.Early apoptosis and cell cycle distribution were assessed by flow cytometry. Results GSAAC significantly inhibited the proliferation, migration and induced the early apoptosis of HepG2 cells;GSAAC blocked HepG2 cells to go to G2 phase from S phase. Conclusion The intrinsic mechanism of GSAAC might relate to the blockage of HepG2 cells to go to G2 phase from S phase thus to inhibit the proliferation, migration and induce the early apoptosis of HepG2 cells.
