Mechanism underlying the apoptosis of L02 cell induced by chitosan-gene nanoparticles

Objective To investigate the mechanism underlying the apoptosis of L02 cell induced by chitosan-gene nanoparticles(CGNP).Methods CGNP were prepared using complex coacervation method.The particle size wag measured using Dynamic Light Scattering System.The morphology of the CGNP Wag observed using momic force microscope(ARM).The nanoparticles at concentrations of5,10,30,and 50 μg/mL(based on the content of DNA)were incubated with L02 cells,respectively.The amount of reactive oxygen species(sos)was determined by fluorescence analysis.The release of cytochrome C(Cyt C)was analyzed by Western-blot.Results The average diameter of the CGNP was 115.6 nm,ranging from 74.14to 136.28 nm.It is demonstrated that the CGNP could induce the release of Cyt C from the mitochondia to the cytoplasm at the concentration higher than 30μg/lnL Conclusion CGNP at higher concentration could induced the injuries ofmitochondria and hence lead to cell apoptosis.
