Regulatory effect of "BUSHENGUCHIWAN" fluid on osteoblast in rats: an in vitro observation.

AIM:To explore the impacts of the concentrated "BUSHENGUCHIWAN"(BSGCW) fluid on proliferation,alkaline phosphatase activity and ultrastructure of osteoblasts.METHODS: Osteoblasts in vitro from cranial bone of Spague-Dawley rats were separated and cultured as the cell model for screening potency of BSGCW.MTT was used to detect the proliferation activity of osteoblasts with the effects of difference concentration of BSGCW fluid at difference time.Differentiating effects of the BSGCW fluid with different concentrations to the cell were evaluated through the examinations of alkaline phosphatase(ALP) activities.Transmission electron microscope was used to study the ultra-structure of cells by the effects of BSGCW fluid.RESULTS: The concentrated "BUSHENGUCHIWAN" fluid showed inhibitory action to the proliferation of osteoblasts in a concentration dependent manner.In the fluid of high concentration(100mg/L) the activity of ALP was decrease significantly(P0.05).There were no obvious differences between low concentration fluid and the control group(P0.05).After action of the concentrated fluid,there were less organelle,such as ribosomes and mitochondria and more vacuole of endoehylema in the experimental group than in the control.CONCUSION:The concentrated BUSHENGUCHIWAN fluid has no significantly promotive effect to the osteoblast metabolism.
