MOLECULAR CHARACTERIZATION OF METHICILLIN RESISTANCE STAPHYLOCOCCUS SAPROPHYTICUS ISOLATED FROM HOSPITAL SAMPLES

The primary cause of antibiotic resistance is genetic mutation in bacteria. The specimens were obtained from different patients under medical attention from Osmania hospital, Hyderabad. The bacterial strains were isolated by serial dilution plate method. Mannitol salt agar is selective for staphylococci because of the high salt concentration. Thus it is confirmed that Staphylococcus saprophyticus isolates developing blaZ gene is responsible for Methicillin resistant which is identified by antibiotic sensitivity test and plasmid isolation. In the present study variable type of resistance was shown against Ampicillin, Vancomicin and sensitivity was shown against Streptomicin, Gentamicin, Tetracyclin, Chloramphenical and intermediate was shown against Rifampicin and Erythromicin. Maximum β-lactamase activity was observed between fractions 33 to 35. The Molecular weight of the purified βlactamase was calculated to be about 41 kDa respectively.
