Determination of blood concentration of vancomycin and norvancomycin by ion-paired HPLC

OBJECTIVE To determine the blood concentration of vancomycin and norvancomycin.METHODS HPLC was used.CH_ 3 CN-ion paired buffer(115∶885,pH3.1) was used as mobile phase on Shimpack C_ 18 (150×6mm,5μm)with column temperature at 40℃. The flow rate was 1mL·min~ -1 . Detection wavelength was 236nm.RESULTS The calibration curve was linear within the range of 0.80~80.19μg·mL~ -1 for norvancomycin and 1.08~108.00μg·mL~ -1 for vancomycin. The average recovery within high and low blood concentration of norvancomycin and vancmycin were 97%~110% with RSD among 2%~6%.CONCLUSION This method was simple,rapid and suitable for the clinical monitor of norvancomycin and vancomycin.
