The research of correlation between 14-3-3β and lympho node metastasis in gastric cancer

Objective To investigate the biological behavior and the clinicopathological significance with silencing of 14-3-3β expression in gastric cancer. Methods RNA interference method was used to knock-down the expression of 14-3-3β in gastric cell lines, and MTT method was used to detect tumor cell proliferation. We used transwell test to check the invasive ability of gastric tumor cells without 14-3-3β expression. Results RNA interference could downregulate the endogenous 14-3-3β expression efficiently in MGC803 and HGC27 cell lines. After transient transfection in HGC27 and MGC803 cell lines. The proliferation rate of transfected cells was significantly decrease compared to the negative control (P values are 0.000, 0.000, 0.000, 0.000, 0.000, 0.001, 0.000, 0.005). Furthermore, the transwell chamber test showed that the absence of 14-3-3β expression in MGC803 could reduce the invasive capability of cancer cells (P values are 0.000, 0.000). Conclusion RNA interference method is effective in koncking-down 14-3-3β expression in gastric carcinoma cell line. 14-3-3β may be involved in regulating cell proliferation and invasion of gastric cancer.
