Investigation of inhibitory effects of 125I-AS on ECV304 cell line and influencing factors of the inhibition.

Objective:To investigate inhibitory effects of 125I-angiostatin(125I-AS)on ECV304 cell line and influencing factors of the inhibition including incubation time,temperature and 125I-AS concentration,etc.Methods:After the extracted AS was labeled with 125I by Ch-T method,the labeling efficiency was measured by paper chromatography.Then Sephadex-G50 column was used to purify the labeled production.The stability and bioactivity of 125I-AS were detected in vitro subsequently.The inhibitory effects of different incubation time,temperature and 125I-AS concentration on ECV304 cell line were detected by MTT method.Results:125I-AS was relatively stable in vitro and its labeling efficiency could achieve to 85%.125I-AS showed obviously higher inhibitory effects on ECV304 cell line than single use of 125I or AS.Moreover,the inhibitory effects increased with the increasing of incubation time,temperature(between 4-37℃) and the concentration.The inhibition reached the highest rate of(79.4±3.7)% at 37℃,96 hours with 370 KBq/ml of 125I-AS(P0.01).Conclusion:125I-AS has good stability and biological activity in vitro.The ECV304 cells can be more evidently inhibited by 125I-AS compared with single use of 125I or AS.Its inhibitions are of time-effect,temperature-effect and dose-effect relationships.
