Study on rapid detection of Vibrio parahaemolyticus by screening method

Objective:To establish a method to enhance the detection capacity and efficiency of Vibrio parahaemolyticus(VP) in seafood.Methods:PCR method was used firstly to do the rapid screening,and then Chinese Standardized method(GB method),to identify the target bacterium.Systematic biochemical specialization was used to final identifying.Results:141 VP strains were screened positive in 356 seafood samples,positive rate was 39.61%.All 141 positive strains were detected by PCR method at a time,while the GB method,115 positive strains were detected by primary culture and 26 others were found by cultured in secondary enrichment medium.The Consistent rate between the biochemical screening method and systematic biochemical specialization method reached 100%.5 strains with TDH virulence was found,while no strain with TRH virulence was detected.Conclusion:The combination of PCR,GB with other typical biochemical method can be used for detecting VP in seafood,which is quick,convenient and effective.The method could be the base for monitoring multi-pathogens pollutant in food.
