The role of MACC1 in the migration and chemoresistance of colorectal cancer

Objective To investigate the functions of metastasis-associated in colon cancer-1(MACC1)in the migration and chemoresistance of colorectal cancer. Methods SiRNA was used to inhibit the expression of MACC1.Transwell assay was performed to study the role of MACC1 in migration.MTT assay was used to detect the chemosensitivity of SW620 cells to 5-Fluorouracil.The relevant protein changes of potential signaling pathways were determined by Western Blot. Results Compared with SW480 cells, SW620 cells showed high expression of MACC1, which could be inhibited by SiRNA transfection.Inhibition of MACC1 by SiRNA significantly suppressed colorectal cancer cell migration.We found that down-regulation of MACC1 sensitized SW620 cells to 5-Fu treatment.Furthermore, the levels of p-ERK and p-Met were also suppressed by inhibition of MACC1. Conclusion Our findings suggest that MACC1 plays an important role in the migration and chemoresistance of colorectal cancer, and may serve as a potential therapeutic target.
