Isolation, enhanced growth, and degradation characterization of a strain marine petroleum degrading bacteria

Abstract In order to systematically explore the growth and biodegradation of marine petroleum degrading bacteria, the growth conditions of a strain marine petroleum degrading bacteria (HDMP2) were optimized. The degradation pathway of diesel by the bacteria was studied. The results showed that the strain of HDMP2 was Halomonas sp.nmyj-1. The main growth factors affecting HDMP2 were rotation rate, inoculum concentration, pH and salt concentration based on the results of variance analysis. When the rotation rate was 179.85 r/min, the inoculum concentration was 2.20 %, the pH value was 7.86, and the salt concentration was 5.99 %, the biomass of HDMP2 was the highest. In addition, when the rotation rate was 180 r/min, the inoculum concentration was 2.73%, the pH value was 7.39, and the salt concentration was 4%, the degradation rate of diesel by HDMP2 was the highest, and the predicted degradation rate could reach 50.31 %. In the degradation process, the degradation of diesel by HDMP2 was a dynamic process. The pathways included surface adsorption, cell uptake and biodegradation. The diesel components were quickly adsorbed on the surface of HDMP2 at the initial stage and most diesel components were finally biodegraded.
