The invention relates to the field of food hygiene and safety, particularly to a bacterial colony total number rapid detection card and an application method thereof. The bacterial colony total number rapid detection card is compose of a lower layer and an upper layer, wherein the lower layer is composed of a first transparent resin layer, a first transparent double-faced adhesive layer, a first gel layer and a ventilation layer which are sequentially arranged from bottom to top; the upper layer is composed of a second gel layer, a second transparent double-faced adhesive layer and a second transparent resin layer; the upper layer and the lower layer are in hinge connection; the ventilation layer is provided with through holes for placing sample solutions; the first gel layer is uniformly mixed with cultivation matrix powder. The application method comprises taking hollow-out blow molding paper as the ventilation layer and is cultivated in an incubator of 35-37 DEG C for 24-28 hours, counting the number of bacterial colonies, and performing conversion to obtain the total number of bacterial colonies in the sample. The structure and the application method of the bacterial colony total number rapid detection card have the advantages that dry powder are uniformly adhered onto transparent double-faced adhesives; the ventilation layer is environmentally friendly in materials and facilitates through hole forming; the transparent resin layers are provided with squares through printing for facilitating number counting. During application, the bacterial colony total number rapid detection card and the application method thereof are rapid in operation, high in sensitivity, accurate in counting and low in cost.
