Tadanafil increases microvascular permeability in mouse brain lymphoma models

Objective To study about Tadalafil increasing microvascular permeability in mouse brain lymphoma models.Methods Comparatively analyze the influence of tadalafil on cerebral microvascular permeability in the mouse brain lymphoma models,by the approaches of using Evan's Blue to test the microvascular permeability,testing concentrations of methotrexate (MTX) by HPLC,and studying ultrastructures by transmission electron microscope.Results The microvascular permeability in tumor-bearing mice treated with Tadalafil were significantly higher than that in the other groups (P < 0.05).The concentration of MTX increased nearly 1.8 times in tumor tissue with Tadalafil pre-treatment.The increment were significantly higher than that in the other groups (P < 0.05).Numerous pinocytotic vesicles in the cytoplasm of the endothelial cells were observed obviously in Tadalafil pre-treated mouse tumor.Conclusion Phosphodiesterase type 5 inhibitor,Tadalafil,could increased the capillary permeability of the tumor tissue in mouse brain lymphoma models.
