Methodological study on the determination of chloramphenicol residues in animal-based food.

2010
[Objective] To establish a method to determinate the chloramphenicol in animal-based food, and to under- stand the pollution condition of chlormycetin residue level among animal-based foods in Shenzhen city, thus prove theory basis for health supervision. [Methods] ELISA was used for screening chloramphenicol in animal-based food and the positive results were proved by HPLC-MS / MS. [Results] The linear range of chloramphenicol in animal-based food by ELISA method was at the range of 0.025-2.0μg / L with linear correlation coefficient of 0.998, and the recoveries were at the range of 63.3%-84.7%, and the limit of detection was 0.02 μg / kg. The linear range of the HPLC-MS / MS method was at the range of 0.1-10.0 μg / L with linear correlation coefficient of 0.999 4. The limit of detection was 0.02 μg / kg. The recovery rate for the spiked samples was at the range of 81.0% to 89.5%. [Conclusion] The ELISA method is high sensitive and simple sample extracting, and suitable for detection for trace chloramphenicol of mounts of sample with lower cost. The HPLC-MS / MS method is suitable for chloramphenicol detection and structure proof with high sensitive and anti-interference.
