The invention discloses a chiral analysis method for nicotine in tealeaves. The chiral analysis method is characterized by comprising the following steps: drying, crushing and sieving a tealeaf sample, adding extraction liquid for extraction and filtration, performing multidimensional gas chromatography-tandem mass spectrometry analysis after automatic solid-phase micro-extraction, and quantitatively detecting the proportion of S-(-)-nicotine and R-(+)-nicotine in the tealeaves by adopting a peak area normalization method. According to the analysis method, the sample can be simply and rapidly treated, the method is low in detection limit and high in sensitivity and accuracy, and the chiral analysis requirement of the nicotine in the tealeaf sample can be met.
