Effect of deoxynivalenol on the mitochondrial membrane potentials and Bax,Bcl-2 expression of BHK-21 cells

To investigate the effect of deoxynivalenol(DON)on the mitochondrial membrane potentials(MMP)and Bax,Bcl-2 expression of hamster kidney cells(BHK-21 cell)in vitro,the change of early apoptosis,mitochondrial membrane potential and apoptosis-related proteins with Bax and Bcl-2 expression were measured by flow cytometry and immunocytochemical staining assay.The results showed that different concentration of DON could induce apoptosis for BHK-21 cells within the experimental concentration and cause apoptosis significantly as compared with control group(P0.05).A marked decrease of mitochondrial membrane potential in a dose and time dependent manner were observed clearly by mitochondrial membrane potentials assay.Immunohistochemical staining analysis showed that,after 24 h treatment with DON,the expression of Bax was significantly higher,while Bcl-2 decreased.These result suggested that DON could induce apoptosis and the MMP disruptionin of BHK-21 cells in vitro.The essential mechanisms of apoptosis might be related to inducing the change of hepatocarcinoma cell cycle,the expression of the Bax increase and Bcl-2 decrease in BHK-21 cells.
