Tissue culture and rapid propagation technique of Capparis masaikai

【Objective】The rapid propagation and culture technique system of Capparis masaikai was established by studying the effects of different disinfectants on explants and different concentrations and combinations of hormone on induction and multiplication of buds. 【Method】Adventitious buds and multiplication were induced using the one-year-old tender shoot. segments and plant as explants in basic MS medium containing different hormone combinations. 【Result】Mercuric-chloride showed better effects than sodium hypochlorite on the disinfection of explants. The sterilization rate was90%, and the explants grew well. The induction number of adventitious buds increased first but fell later with the increase of 6-BA concentration in induction medium of bud. The germination rate was the highest when 6-BA concentration was 1.0mg/L, the induction rate was higher when NAA was 0.5 mg/L. When 6-BA concentration was greater than NAA concentration, multiplication coefficient was lower, and when 6-BA concentration was less than NAA concentration, multiplication coefficient was higher with the maximum of 3.8. The cluster buds grew better. The plantlets were thick, and the leaf color was normal. 【Conclusion】The sterilization effect of mercuric chloride was better than that of sodium hypochlorite. The optimal medium recipe for inducing buds and multiple shoots of Capparis masaikai was MS+1.0 mg / L 6-BA+0.5 mg / L NAA and MS+0.5 mg / L 6-BA+1.0 mg / L NAA, respectively.
