Screening and identification of dominant allergens of Dermatophagoides pteronyssinus and Dermatophagoides farina.

Objective To screen and identify the dominant allergens relative to Chinese in house-dust mites Methods With SDS-PAGE,Western blotting and LC-MS/MS methods,the dominant allergens have been identified and analyzed from extracts of Dermatophagoides pteronyssinus(D.p) and Dermatophagoides farina(D.f).Results There were some unique protein bands in quantity and molecular size among three samples of D.p and D.f.Composition of proteins had more differences between D.p and D.f samples,less differences between two D.f samples.Molecular size of the dominant allergens(protein) of D.p were about 12~15kDa and 94~98kDa,D.f about 49~53kDa and 94~98kDa.In Western blotting,a D.f sample had 49~53kDa and 94~98kDa allergens reaction bands,but in another sample they could not be seen.With LC-MS/MS methods,64 allergen proteins of D.p and 84 allergen proteins of D.f were identified from dominant allergen bands,including some house -dust mite allergens reported,many kinds of enzymes,translation elongation factor,ribosomal protein,outer membrane(lipo -)protein,keratin,glycine cleavage system protein,binding protein,heat shock protein,regulation protein,hypothetical protein,pol polyprotein and pol protein etc.Conclusion There are differences in composition and immunoreactivity of dominant allergens of D.p and D.f.The dominant allergens relative to Chinese in D.p and D.f may include Der p 2(group 2 allergen of Dermatophagoides pteronyssinus),Der p 14,Der f 3,Der f 15 and Der f 17 et al.Some proteins mentioned above from D.p and D.f may be potential allergens.
