Vitamin B12,one of the vitamin B complex,plays an important role inenergy metabolism and also acts as catalysts in protein synthesis.Some eliteathletes also supply vitamin B12 as a nutritional ergogenic aid.Since the goodquality is essential,an analyze method to determine the amount of vitamin B12 inpharmaceutical preparations is necessary.A rapid,simple,and reliablereversed-phase high performance liquid chromatographic(HPLC)method wasdeveloped for the assay of cyanocobalamin(Vit B12)in bulk drug andpharmaceutical preparations.Cyanocobalamin and other water-soluble vitaminscan be also separated.The samples was analyzed on an Alltima C18(25 cm×4.6mm I.D.;particle size,5μm)column with a mobile phase of 25 mM potassiumdihydrogen phosphate/acetonitile(V/V=86/14)at a flow-rate of 1.0ml/minand detection at 210nm.Cephazolin was used as an internal standard.Theretention times of cyanocobalamin and cephazolin were 8.9 and 7.7 minutes.Anequation was presented for linear relationship between peak height ratios ofcyanocobalamin to cephazolin and the cyanocobalamin concentration over arange of 0.15-0.75μg/ml(r=0.999).Standard addition recoveries were greaterthan 96% with four pharmaceutical preparations.Furthermore,paired assayresults by microbiological medium diffusion assay and described HPLC methodswere obtained from bulk drug,500μg and 100μg capsule,62.5μg tablet,1000μg injection,containing cyanocobalamin.Statistical tests were used to assess theequivalence of the paired assay results.At the 95% confidence level,nosignificant intermethod differences were noted for the assay results for each pair.The results indicated the proposed HPLC method is suitable substitute for themicrobiological method of the United States Pharmacopeia for potency assays ofcyanocobalamin preparations.The analytical method can be used to determinethe commercial vitamin B12 preparations.It can help athletes to choose the goodquality of vitamin B12 and achieve the best supplementation.
