Effects of Temperature, Sucrose and Boric Acid on Vitro Pollen Germination of Jatropha curcas

The experiment was conducted with the fresh pollens of 4-year-old Jatropha curcas by in vitro culture to study the effects of culture medium composition and temperature on pollen germination.The results showed that in single factorial experiments,sucrose and boric acid could stimulate pollen germination in certain concentration range,they could inhibit the germination at higher concentrations.The optimal concentrations of sucrose and boric acid were 10% and 0.03% respectively,and the pollen germination rate was up to 23.6%;The optimal temperature was 30℃,and the pollen germination rate was up to 20.2%.In orthogonal design experiment,the effects of sucrose,boric acid and temperature on the pollen gernmination were extremely remarkable,and the optimal culture medium was 12% sucrose +0.01% boric acid at 33℃,in which the rate of pollen germination was up to 36.1%.
