The invention discloses a reagent for detecting R46S mutation of a Pygo2 gene in a Wnt signal channel based on a peptide nucleic acid probe, a PCR detection method and application. The detection reagent comprises primers, peptide nucleic acid fluorescent probes and wild type complementary peptide nucleic acid sequences, wherein forward primers are shown in SEQ ID NO.1, NO.3, NO.5 and NO.7 in a sequence table; reverse primers are shown in SEQ ID NO.2, NO.4, NO.6 and NO.8 in the sequence table; peptide nucleic acid fluorescent probes are shown in SEQ ID NO.9, NO.10, NO.11 and NO.12 in the sequence table; and the wild type complementary peptide nucleic acid sequences are shown in SEQ ID NO.13, NO.14, NO.15 and NO.16 in the sequence table. The detection reagent disclosed by the invention can rapidly find mutation condition of the Pygo2 gene in the Wnt signal channel from a transcriptional level, and a tool is provided for anti-cancer drug screening, discussion of new targeted drugs and basic scientific research.
