Construction and study of the immunogenicity of eukaryotic plasmids of the sap2 gene of Candida albicans.

Objective To construct eukaryotic expression plasmids of the sap2 gene of Candida albicans and analyze their immunogenicity.Methods Using PCR,the sap2 gene was amplified from standard strain ATCC 64550 and inserted into eukaryotic expression vector pcDNA3.1.Eukaryotic expression plasmids were isolated and injected into BALB/c mice.Antibodies against sap2 were detected by ELISA and cellular immune response was detected by flow cytometry.Results The gene sap2(1 197 bp) was harvested by RT-PCR.Mice were immunized with the pcDNA3.1-sap2 constructed and were induced to have a specific anti-sap2 IgG titer of 1∶1 600 and increased percentage of CD4+T and CD8+T cells.Conclusion The gene sap2 of C.albicans was harvested.The eukaryotic expression plasmid pcDNA3.1-sap2 can induce cellular immune response and humoral immune response in BALB/c mice.
