Effects of total flavonoids of Eucommiabark on the proliferation of rat osteoblast and synthesis of I collagen protein

Objective To investigate the effects of total flavonoids of Eucommia bark on proliferation of rat osteoblast and synthesis of collagen protein.Methods The osteoblast was isolated from the crinial bone of Sprague-Dawley rats at postnatal day 2,identified by alkaline phosphatase staining and cultured with 10,100,200 and 400 μg·mL-1 total flavonoids of Eucommia bark.The proliferation of cells was identificated by ELISA using MTT,and the synthesis of Ⅰ collagen protein was detected by confocal laser scanning microscopy.Results The total flavonoids of Eucommia bark at the concentration from 10 μg·mL-1 to 200 μg·mL-1 improved evidently the growth and proliferation of rat osteoblast but didn't have obvious promoting effect to the synthesis of Ⅰ collagen protein.Conclusion The flavonoids of Eucommia bark are useful for the supporting of bone through promoting the quantity of osteoblasts.
