The expressions of STAT1 and PDGF in lung tissue of pulmonary fibrosis rats induced by bleomycin

2008
Objectives To study the expressions of STAT1 and PDGF in lung tissue of pulmonary fibrosis model induced by bleomycin in rats, and discuss the possible mechanism. Methods Thirty Wistar rats were randomly divided into two groups(15 in each group),they were bleomycin group(BLM group) and control group(NS group).①The BLM group was intratracheally instilled with belomycin(BLM)(5mg/kg);② NS group was intratracheally instilled with saline(5mg/kg).Five rats in each group were sacrificed at 7,14,and 28 days after intratrachel instillation. The bronchoalveolar lavage fluid(BALF) of right lung was gotten and the cells counting as well as differentiation was figured out.Lung expressions of STAT1 and PDGF protein were assessed by immunohistochemistry. Results The expressions of STAT1 and PDGF protein in BLM group were the highest at 7th and still elevated over NS group at day 28.The expressions were correlated with the counting of cells,respectively(r=0.892 and 0.902,P0.01).Conclusion The hyperexpressions of STAT1 and PDGF protein in lung may be the mechanism of pulmonary fibrosis.
