The immobilization of Candida antarctica lipase B and its application in synthesis of vitamin A palmitate

Three kinds of resin with good immobilization efficiency(ECR1030M, LX-1000 EP and MC150EP) were selected to optimize the immobilization conditions of Candida antarctica lipase B.The optimal parameters of physical adsorption for immobilization were as follows: immobilization medium disodium hydrogen phosphate-citric acid buffer with p H 8.0 or 6.0, ratio of lipase solution(18 g/L) and resin 20 ∶1(V/W), incubation temperature 30 ℃, speed 180 r/min, and incubation time 3 h. The specific enzyme activity of the immobilized lipase were 1 055.7 U/g(ECR1030M),1 077.0 U/g(LX-1000EP), and 1 024.3 U/g(MC150EP), respectively. The enzymatic properties of the immobilized lipase were further studied and the immobilized lipase on ECR1030 M showed best operation stability. After using for 7 batches, the immobilized lipase retained over 90% of its activity, and the yield of vitamin A palmitate reached above 85%.
