Morphogenesis from cultured juvenile explants of Murraya koenigh spreng

Plantlets were obtained in vitro from immature embryos of Murraya koenigii Spreng. on media with activated charcoal. Axillary shoots were induced on nodal explants on SH medium by 2-iP (5mg/l) and IBA (0.5mg/l). Employing MS medium with 0.25% activated charcoal, it was possible to induce axillary shoots on a) nodal explants by i) BAP (5mg/l) or ii)2,4-D (5mg/l), NAA (1mg/l) and 10% coconut milk or by iii) NAA (3mg/l) and BAP (0.3mg/l), b) cotyledonary nodes by NAA (1mg/l) and c) epicotyledonary nodes by NAA (1mg/l) and 6% sucrose. Adventitious shoots could be induced on hypocotyl region of explants of embryonic axis by NAA (1mg/l). Rooting in vitro could be induced with IBA (2mg/l) and 4% sucrose. On transfer to medium with 7% sucrose there was rapid elongation of roots. Somatic embryogenesis was induced in the stem region of shoot explants cultured horizontally in media with 2,4-D (5mg/l), NAA (1mg/l) and 15% coconut milk. Anatomical studies documented the presence of globular and heart-shaped somatic embryos and their superficial origin.
