Expression of hirudine by transformed yeast and anticoagulating agent composition

PROBLEM TO BE SOLVED: To provide the subject new yeast transformed with a plasmid containing a DNA sequence coding for hirudines, a leader sequence secreting genetic product and a DNA sequence containing a transfer signal of gene, etc., and useful for the production of anticoagulating substance, etc. SOLUTION: The objective yeast is transformed by a functional DNA block integrated into a plasmid containing a replication initiation point in the yeast or integrated in a chromosome of the yeast. The block at least contains a DNA sequence (H gene) coding for hirudine, its derivative or precursor, a leader sequence(Lex) containing elements necessary for achieving the secretion of genetic product and a DNA sequence(Str) containing a signal for the transfer of the H gene in the yeast. The yeast is useful e.g. for the production of hirudine compounds having anticoagulating action. The yeast can be produced by introducing a functional DNA block containing H gene, Lex and Str into a yeast of the genus Saccharomyces and transforming the yeast. COPYRIGHT: (C)1997,JPO
