Purification of Red-crowned crane serum IgY and production of antiserum against the IgY.

The immunoglobulin Y in serum of Red-crown crane was crudely extracted through saturated ammonium sulphate method,and purified by Ion-Exchange Chromatography with cellulose DEAE-52.Furthermore,purity of the immunoglobulin was identified by SDS-PAGE.The polyclonal rabbit anti-red-crowned crane IgY antiserum was obtained according to standard immunization procedure.The result showed that the purified IgY displayed 63 ku and 27 ku protein tope in SDS-PAGE analysis and its purity was higher than 90%.The polyclonal rabbit anti-red-crowned crane IgY antiserum was produced and the titer of antiserum was up to 1∶32 by agarose Double Immunodiffusion.These results could contribute to the establishment of Red-crowned crane serum monitoring system
