Expression of special AT rich sequence binding protein-1 in lung adenocarcinoma and its effect on proliferation, invasion and apoptosis of lung adenocarcinoma cells

Objective The study aimed to explore the relationship between the expression of special AT rich sequence binding protein-1 (SATB1) and clinical characteristics in lung adenocarcinoma and its role in the proliferation, invasion and apoptosis of lung adenocarcinoma cell line A549. Methods Tissue sections were stained by immunohistochemistry streptavid-in-peroxidase (SP) method. The green fluorescent protein expression vector of SATB1-small interfering RNA (siRNA) was constructed and transfected into lung adenocarcinoma cell line A549. The effect of transfection was observed by fluor escence microscope. The silencing efficiency was measured by Western blotting. Cell proliferation, invasion and migration activity were detected by methyl thiazol tetrazolium (MTT), Transwell and wound-healing assay. Apoptosis rate was measured by flow cytometry. MTT, transwell and wound-healing assay were adopted to study cell proliferation, invasion and migration activity, and the apoptosis rate was tested by flow cytometry. Results SATB1 was highly expressed in human lung adenocarcinoma tissues (positive rate 61%), and was lowly expressed in adjacent tissues (positive rate 20%), and the expression level was negatively correlated with the degree of differentiation (P<0.01). After transfection with SATB1-siRNA, the transfection efficiency was about 70%, and the expression of SATB1 in A549 cells was decreased (P<0.01). Compared with the negative control group, the cell proliferation, invasion and migration ability were significantly decreased (P<0.01). The apoptotic rate was increased (P<0.01). Conclusion SATB1 is closely related to the pathogenesis and development of lung adenocarcinoma, which may provide important clues for finding potential therapeutic targets and prognosis of lung adenocarcinoma.
