Study on callus induction and tissue culture of Taxus cuspidata

Inducing frequency of calli from young stems of cutting plantlets of Taxus cuspidata as explants on the different media was 86.7%(B 5-B),60.2%(MS-A)and 37.0%(MS-B),respectively.On the MS medium with 2,4-D and NAA,the inducing frequency of call was 1.5～2 times higher than that on the MS medium only with 2,4-D.Results also showed that MS medium was good for calli to grow on.The growth cycles of calli reduced from 35～40 days to 15～18 days during subculture.There were some differences of inducing frequency and growth rate of calli from young stems between trees and cutting plantlets,between the trees and even from different branches of the same trees.
