Influence of the new synthesis of chalcones derivative L6H21 on CD4+/CD8+T cell ratio at acute rejection stage following rat limb allograft

Objective To explore the influence of the new synthesis of chalcones derivative L6H21 on CD4+/CD8+T cell ratio at acute rejection stage following rat limb allograft. Methods The hind limbs of 36 healthy male SD rats were served as donors, and 72 healthy male Wistar rats were used as recipients to establish the rat model of hind limb allograft. All the rat models were randomly divided into three groups according to the different postoperative immunosuppressive regimes: 0.9% NaCl control group (group A), cyclosporine A group (group B), chalcone derivative L6H21 group (group C). At 1, 3, 5 and 7 days after the allograft transplantation, appearance changes and rejection occurrence time of the transplanted limbs from each group were observed. CD4+ and CD8+T cells were sorted by flow cytometry in order to figure out the changes of CD4+/CD8+T cell ratio. Results The average survival time was (6.00±0.67) d for group A, (23.62±1.21) d for group B and (25.34±1.70) d for group C. At 1 day after the surgery, no CD4+/CD8+T cell ratio difference was found among all the groups. At 3 days postoperatively, CD4+/CD8+T cell ratio was obviously increased in group A. Compared with the other two groups, the difference was statistically significant (P 0.05) . After 5 days postoperatively, CD4+/CD8+T cell ratio was decreased in all the groups, and group B and C reduced more significantly (P<0.05). Conclusion The new synthesis of chalcones derivative L6H21 can reduce CD4+/CD8+T cell ratio at acute rejection stage following rat limb allograft, and prolong the survival time of rat limb.
