Eucalyptus 12ABL callus induction and plantlet regeneration propagation system construction

The hypocotyls of Eucalyptus 12ABL were used as explants, and a rap established. A systematic research was conducted on obtaining the calli wi micropropagation system was shoot organic potentiality, adventitious buds differentiation, multiplication, and rooting culture. The results showed that the optimal medium for inducing calli was modified H medium ＋0.5 mg/L 6-BA ＋ 0.5 mg/L NAA ＋ 40 g/L sucrose, and for inducing adventitious buds modified H medium ＋ 1 mg/L 6-BA ＋ 0.1 mg/L NAA ＋ 40 g/L sucrose, with the inducing rate reaching 65.7 % and 54 % respectively. The optimal multiplication medium was MS＋I mg/L 6-BA ＋ 0.1 mg/L NAA ＋ 30 g/L sucrose, with a propagation coefficient being 3.4. And the optimal rooting medium was 1/2 MS＋ 0.5 mg/L IBA ＋ 30 g/L sucrose, with the mean rooting rate being 100 % and root number 3.6 roots/plant.
