Monocyte Chemotactic Peptide-1 (Mcp-1) and NAP-1/IL-8 Formation in Baboon Septicemia are Partially TNF Dependent

Cytokines like IL-1 and IL-6 are released in sepsis in a TNF dependent manner. Therefore we asked the question, whether TNF might be responsible for MCP-1 and NAP-1 production in baboon septicemia (2 hours infusion of live E. coli 2 x 109 CFU/kg). We performed sepsis experiments with and without pretreatment with humanized TNF-AB (CDP571, Celltech) 1 mg/kg, which completely neutralizes circulating TNF. MCP-1 was increased from 0.6 ng/ml at baseline up to 6.8 ng/ml at 4 hours.
